The aim of the proposed project is to characterize interspecific physiological differences in selected lichen photobionts (Trebouxia genus) with different type of chloroplast morphology. Within the experiments, techniques of cultivation of algae in crossed gradients of temperature and light will be used. A growth and production of algal colonies will be analyzed periodically within the experiments by an analysis of photogrammetrical images, determination of chlorophylls and carotenoids contents and by a weighing of algal colonies. The aim is to assess the effects of limiting conditions of temperature, light and presence of heavy metals. The effect of these factors on primary photochemical processes in photosystem II will be studies by the parameters of chlorophyll fluorescence in vivo. The results of the project will contribute to clarify physiological response of lichen photobionts to optimal and limiting conditions of temperature, light and heavy metals. Analysis of physiological processes at the cellular level of Trebouxia photobiont culture will elucidate the effect of proportion of individual reproductive stages within a lichen thalus on heterogeneity of photosynthetic production.
