Description | Analysis of total DNA content by flow cytometry and of noncoding chloroplast DNA using restriction enzymes indicated DNA variability in Poa pratensis agg. within the Czech Republic. Total DNA content was measured for 52 plants from seventeen locations in the Czech Republic, primarily from Moravia. The greatest DNA content was found in 7 plants from the Chrudim area. Using a standard (a plant with a known amount of DNA and precise chromosome count) the amount of nuclear DNA was converted to chromosome number, which spanned from 68 to 81 chromosomes. This method was used for all measurements. 14 plants from the Brno area had (according to this conversion) 31 to 56 chromosomes. Two plants from the Moravian karst had 32 chromosomes. Plants from south of Brno all had low amounts of nuclear DNA, from 30 to 45 chromosomes. It is possible to consider that the Panonian representatives of Poa pratensis agg. contain less nuclear DNA than those of other areas. Variability was also studied using the noncoding trnL intron and trnL-F intergenic spacer from the chloroplast. Restriction analysis was carried out using Alu I, BsaJ I, and Bsm I. Three cultivars from the Plant Breeding Station Hladké Životice were used as models. Three types of sequence, labeled MM, MS and HM were found within them. In 120 samples from Europe, North America and Asia these three types were found as well as one additional type. The total number of analyzed plants from the Czech Republic was 23, of which type MS was found in plants from the Brno area, one from Chrudim, 2 from South Bohemia, from the Moravian Karst and from Southern Moravia. Type MM was found in one plant from Ostopovice u Brna and one from the Chrudim area. Type HM was found in only one plant from Ivančice u Brna. It is interesting that in such a small area, nearly all the types found in various places in the Northern Hemisphere were found. No relationship was found between the type of chloroplast DNA and the amount of nuclear DNA for a given plant. |
