Popis Human embryonic stem cells are able to differentiate into every cell type in the human body making them a tremendous cell source for regenerative medicine. However, the production of human embryonic stem cell lines for clinical use is challenging as clean rooms, highly-qualified personnel, standard operating procedures for both manufacture and quality control are required. Here we present the derivation of clinical-grade hESC lines in cooperation with the Centre of assisted reproduction - University Hospital Brno which provided 6-day old blastocysts. Laminin 521 in combination with Nutristem, Human serum albumin, and ECadherin was used for the mechanical derivation. Human embryonic stem cells were cultured on laminin 521 in a Nutristem medium. The derivation itself and subsequent culture are xeno-free, feeder-free, and fulfill standards of current good manufacturing practices. In-depth quality control provides essential information about the safety, pluripotency, and differentiation potential of lines. Presented clinical-grade hESC lines were established according to state-of-the-art technology that makes them excellent cell source for stem cell-based therapies.
