The project proposes a new development of single-molecule immunoassays using the luminescence of photon-upconversion nanoparticles. This type of luminescence is characterized by excitation in the infrared region and emission of short-wavelength radiation (mostly visible). The low background of this type of luminescence simplifies the imaging and counting of individual nanoparticles; when used as a luminescent label, the counting of the single analyte molecules is possible. The innovation is based on using microfluidic chips for automation. Microfluidics decreases analysis time, reagent consumption, detection limits, and facilitates the analysis of microscopic samples (single-cell analysis). The methodology will be verified by the detection of known protein biomarkers: (A) prostate-specific antigen in blood serum and plasma and (B) protein biomarker p53 at the level of individual cells. The project aims to improve disease diagnostics and monitoring the course of disease treatment and advances the toolbox of cell and molecular biology.
