Description Cytokinins (CK) have been implicated in regulation of many aspects of plant growth and development. Recent genetic studies suggest that CK play distinct and sometimes even opposite roles in the regulation of shoot and root development in Arabidopsis. However, the underlying molecular mechanisms are still rather elusive. Here we present results of a comprehensive approach in the study of differential shoot and root CK response. To study immediate-early and delayed CK response, six days old seedlings of Arabidopsis th.(Col-0) were treated with 5 uM 6-benzylamino purine for 30 min and 2 hours, respectively. Root and shoot tissue was analysed separately for both studied intervals. The proteome analysis was performed using 2-D gel electrophoresis and luminescent SYPRO Ruby gel staining. Proteins in the selected spots were identified using LC-MS/MS. Applying that approach allowed us to identify a number of unique, qualitatively and/or quantitatively different proteins in the root or shoot samples within both studied intervals.
