Description | Purine bases represent the basic constituents of DNA, their analysis is useful in the diagnosis of several diseases and metabolite disorders. The related bases, such as 6-aminopurine (6-AP, 2-aminopurine (2-AP) and 2,6-diaminopurine (2,6-DAP) were studies by spectroscopic and RP-HPLC techniques. Studied APs can be separated based on their spectral properties. Elution order related to polarity of molecule, in MF MetOH/30mM phosphate buffer (15:85), was following: 2,6-DAP, 2-AP, 6-AP. The phosphate concentration is not crucial. Experimental pKa values of model APs were calculated from corrected capacity factor dependence of APs on pH. The basicity increasing in order 2-AP<6-AP<2,6-DAP. Studies of basic structures will be used for nucleosides and nucleotides study, where 2-AP and 2,6-DAP can be used as fluorescent labels. |
