The application of MALDI MS whole-cell profiles of the genus Pseudomonas

Description The genus Pseudomonas is wide-spread in the enviroment, food and human and veterinary clinical material. Pseudomonads are often resistant to antibiotics and chemicals and take part in structure of biofilms with the posibility of colonization of different mucosa and tissue. They are responsible for nosocomial and systemic infections. Their importance lies in the potencial of enzymatic ways of biodegradation and geochemical cycles too. The genus Pseudomonas sensu stricto a poddruhů. comprises more than hundered validly described species and subspecies, at the present. Classification of environmental isolates is often complicated due to the heterogeneity and genetic variability of the species. The evolving taxonomy of the genus is in great progress now. Matrix-assisted laser desorption/ionization time of flight mass spectrometry is an analytical technic, which identifies specific markers of the bacterial proteome. This method has been succesfully applied to identification of related genus Aeromonas and is able to distinguish two closely related organisms, that are misidentified phenotypically or genotypically. In our work, sample preparation procedures were optimised including culture preparation and conditions of MALDI-MS analysis of 90 proteobacteria strains. By the use of the method MALDI TOF there were obtained clear and rapid outcomes of mass spectra of markers. The data processing means the identification and quantification of peak list, the calculation of the similarity among spectra, cluster analysis and finally classification of the sample. There was acquired typical mass spectra of characteristic MALDI MS profiles which enable rapid characterization of the microorganisms on the the genus, species and strain levels.
