Description The RAW 246.7 murine leukaemic monocyte/macrophage cell line possesses the ability to differentiate into different cell types, such as osteoclasts in RANKL and M-CSF dependent way or dendritic cells (by bacterial lipopolysaccharide). In this process, transcription factors c-Jun and c-Fos are necessary. Integrin beta 3 is important for function of osteoclasts as well by mediating adhesion to the bone surface and the bone matrix resorption. The promotor of integrin beta 3 gene contains binding sites for c-Jun and c-Fos transcription factors as described in in HD11 myeloblasts. Therefore, it is possible that crosstalk between these proteins exists and is important in regulation in osteoclasts activity or differentiation. These molecules can be important in dendritic cell differentiation as well. We studied the expression of c-jun, c-fos a itgB3 during the LPS or M-CSF-RANKL driven differentiation of RAW cells by using quantitative RT-PCR and western-blotting. We will link our results to a functional interactions analysis.
