Description | Method for the laboratory diagnosis of American foulbrood was developed and optimized. The analysis is based on the detection of the bacterium Paenibacillus larvae, the prepared anti-P. larvae antibody is used for the specific capture of the bacterium in an immunoassay using photon-upconversion nanoparticles (UCNPs) as a label. The UCNPs were conjugated with antibody or streptavidin using a linker based on polyethyleneglycol; this detection element was used as an alternative to conventional labeling in enzyme-linked immunosorbent assay (ELISA). The upconversion-linked immunosorbent assay (ULISA) based on UCNPs provides a detection limit of 2.9×10^3 CFU/mL, representing a 22-fold improvement compared to ELISA. The assay was successfully applied to analyze real samples of bees, larvae, and bottom hive debris. The achieved results demonstrate the suitability of the method for laboratory diagnosis of American foulbrood. |
